大约 2% 至 3% 的男性和 6% 至 7% 的女性患有严重抑郁症。现有药物只能部分缓解大约 40% 的患者症状。大多数抗抑郁药物都是基于已有 50 至 60 年历史的理论,该行业迫切需要新的药物开发目标。近十年来,在动物模型中,许多基因与抑郁症有关,而且严重的抑郁症确实在人类家族中存在,这表明遗传和环境因素都有影响。根据早期研究,抑郁症与血清素信号基因(包括 p11、SERT 等)的功能障碍有关。在某些情况下,抑郁症的基因疗法被发现相对安全,尽管事实上它可能看起来比药物更具风险和更具侵入性。因此,关于永久治疗重度抑郁症的这些基因的最安全传递机制的领域正在不断发展。因此,本综述总结了导致抑郁症的各种基因的传递机制及其分子机制和细胞水平的传递。
