摘要
近年来,科学界一直试图通过统一和整体的方法来解决不同的疾病,这种方法是基于这样一种概念,即在一个全面的总体方案下可能针对明显不同的疾病。换句话说,各种不同的疾病被归类在“构象疾病”的标签下,因为每种疾病的触发原因都是一种特定蛋白质的错误折叠,这种蛋白质的不平衡和积累导致了每种不同疾病特有的所有其他下坡生物分子事件。与此同时,研究蛋白质错误折叠和积累的分析技术也得到了发展,从而为构象疾病的研究提供了有效的技术支持。在这种情况下,表面等离子体共振(SPR)广泛地应用于与构象疾病相关的许多不同方面的研究,提供了实时调查、使用少量生物材料和模拟细胞环境的可能性,而无需重复使用荧光标记。在这篇综述中,在简要介绍构象疾病和SPR技术之后,对SPR的各种用途进行了全面的描述,以研究这些疾病中涉及的生物分子机制,以便为读者提供一个详尽的清单,以及SPR在此类主题中使用的关键观点。阿尔茨海默病的案例在更深层次上进行了讨论。我们希望这项工作将使读者意识到所有可能的SPR实验方法,这些方法可用于开发新的可能的治疗策略来治疗构象疾病。
