利用网络药理学探索益精汤治疗多囊卵巢综合征的机制

摘要

背景:据报道,益精汤 (YJD) 是一种中药方剂,可有效治疗多囊卵巢综合征 (PCOS)。然而,益精汤治疗 多囊卵巢综合征的潜在机制仍不清楚。 目的:在目前的工作中,系统地分析益精汤的有效成分及其治疗多囊卵巢综合征的机制。 方法:从公共数据库中筛选养生汤的有效成分和多囊卵巢综合征的治疗靶点。采用网络药理学方法分析养生汤治疗多囊卵巢综合征的成分、潜在靶点和通路。 结果:从益精汤中鉴定出 103 种活性成分,其中 82 种被 65 个与 PCOS 相关的靶点击中。通过构建疾病共同靶标化合物网络,筛选出五种成分(槲皮素、花生四烯酸、β-谷甾醇、β-胡萝卜素和胆固醇)作为养生汤的关键成分,可与10个中枢基因(VEGFA、AKT1、TP53、 ALB、TNF、PIK3CA、IGF1、INS、IL1B、PTEN)对抗 PCOS。这些基因主要参与前列腺癌、类固醇激素生物合成和EGFR酪氨酸激酶抑制剂耐药通路。此外,分子对接结果表明,益精汤的成分与hub基因具有良好的结合亲和力。 结论:这些结果表明养和汤通过调节雄激素和胰岛素水平,改善炎症微环境来治疗多囊卵巢综合征。
