亲环素A (CypA)是一种普遍存在且高度保守的蛋白质。CypA是免疫抑制剂环孢霉素A (CsA)的细胞内靶蛋白,通过肽基脯氨酰顺反异构酶(PPIase)发挥重要的细胞作用。越来越多的研究表明,CypA在多种人类癌症中上调。除了参与多种肿瘤的发生和发展外,CypA的过表达也被证明与恶性转化密切相关。手术、化疗和放疗是癌症的三种主要治疗方法。化疗和放疗经常被用作癌症的直接或辅助治疗。然而,各种副作用和对化疗和放疗的耐药性给这两种治疗形式带来了巨大的挑战。根据最近的报告,CypA可以改善癌症的化学敏感性和/或放射敏感性,可能通过影响耐药相关蛋白的表达、细胞周期停滞和丝裂原活化蛋白激酶(MAPK)信号通路的激活。在这篇综述中,我们着重于CypA在癌症中的作用,它对癌症化疗和放疗敏感性的影响,以及作用机制。提示CypA可能是癌症化疗和/或放疗的新的潜在治疗靶点。
