简介:胃癌是一种众所周知的恶性肿瘤,每年导致全球数百万人死亡。由于缺乏针对胃癌的特异性生物标志物,大多数患者在诊断时已处于疾病晚期,导致预后不良和死亡率较高。因此,迫切需要新的生物标志物来进行早期诊断并提高生存率。 方法:在本研究中,我们对 21 名胃癌患者的肿瘤样本进行了 RNA 测序。共鉴定出3192个差异表达基因(1589个上调基因和1603个下调基因)。随后,我们应用文本挖掘算法进一步分析这些数据,并选择 30 个代表性基因作为胃癌新型生物标志物的候选基因进行研究。 结果:在这些基因中,基于对 134 个样本进行的蛋白质印迹和免疫化学验证,我们确认瞬时受体电位褪黑素 8 信道 (TRPM8) 是一种新型生物标志物。与正常胃组织相比,肿瘤组织中TRPM8的表达水平显着升高。 结论:本研究深入了解 TRPM8 在细胞增殖中的潜在作用。此外,TRPM8可能作为胃癌患者的潜在治疗靶点。
